Head coach Sean McDermott announced Friday on WGR 550 radio in Buffalo that Rapp will miss the game because of a hip injury. Rapp has not participated in practice this week.
Cole Bishop and Cam Lewis could receive more playing time in the absence of Rapp, who was injured in the second quarter of Sunday's divisional playoff win over the Baltimore Ravens. Rapp was ruled out midway through the third quarter of the Bills' 27-25 victory.
Rapp, 27, recorded three tackles and an interception in the two playoff games.
A Super Bowl champion with the Los Angeles Rams, Rapp had 82 tackles, two interceptions and a forced fumble this season.
McDermott said Friday morning that linebacker Matt Milano (hamstring) and cornerback Taron Johnson (neck, shoulder) were expected to be full participants in practice later that day.
