The shutout was the second of the season for Quick, the 60th of his career and his first against Pittsburgh.
Alexis Lafreniere scored for the Rangers, who had their 11-game point streak broken in their previous game, against Dallas. New York improved to 8-2-1 on the road.
Quick hadn't been dominant against the Penguins, going 5-4-5 in his career against them prior to Wednesday.
The Penguins have lost three of their past four games. They were shut out for the first time this season.
Pittsburgh goaltender Tristan Jarry kept his team in it, making 35 saves.
It was the first meeting of the season for the Metropolitan Division rivals. The Rangers are 6-1-1 in the teams' past eight meetings.
New York's Artemi Panarin, who had his 15-game point streak snapped against Dallas, now has gone two games in a row without a point after having at least one in every game prior.
The Penguins had several good chances early in the first period but could not convert.
New York then took over much of the momentum.
At 5:10 of the first, Lafreniere gave the Rangers a 1-0 lead. After Pittsburgh defenseman Ryan Graves turned the puck over in the neutral zone, Mika Zibanejad sprang Lafreniere on a breakaway just ahead of a couple chasing Penguins players.
Lafreniere lifted a backhander over Jarry's glove.
Pittsburgh thought it had a tying goal at 15:30 of the first during a power play. Quick lost his glove when he got bumped by the Penguins' Radim Zohorna. Play continued, and Pittsburgh took advantage of the situation when Vinnie Hinostroza beat Quick on the glove side from the left circle.
However, New York successfully challenged that the Penguins were offside, negating the goal.
Jarry kept the Penguins in the game in the second, turning away 17 shots. The Rangers led in shots, 32-17, going into the third.
