Busch had two hits and two RBIs, Kyle Tucker notched two RBIs and Dansby Swanson posted two hits for the Cubs, who have won five of their last seven.
Corbin Carroll had three hits, including a leadoff homer to extend his hitting streak to 12 games for the Diamondbacks. He has reached base in all 21 games this season and 27 straight dating to last September.
Suzuki homered for the second straight game, and the Cubs have seven homers in the first two games of the series.
Cubs starter Ben Brown gave up one run and three hits in four innings, with four walks and six strikeouts. Four relievers combined to retire 14 straight before Tim Tawa's two-out homer in the ninth.
Caleb Thielbar (1-0) retired all six Diamondbacks he faced after replacing Brown.
Arizona starter Zac Gallen (1-3) gave up six runs and six hits in the first two innings, using 64 pitches, before settling down to retire 13 of the last 14 he faced. He had five strikeouts and two walks.
Carroll hit Brown's third pitch of the game - a 2-0 fastball - for a 426-foot homer, his seventh of the season and fourth in the last 10 games. He is 22-for-54 (.407) with 10 extra-base hits and 13 RBIs in the last 12 games.
The next two batters reached on first baseman Busch's fielding error and a walk, but Brown halted the threat by inducing a soft line drive followed by two strikeouts.
The Cubs responded quickly. Ian Happ walked and scored on Tucker's line drive off the right field wall for a triple. Busch's one-out single made it 2-1.
Miguel Amaya walked and Jon Berti singled to open the Cubs' four-run second. Amaya took third on a lineout and scored on Tucker's groundout off Gallen's glove for the second run before Suzuki hit a 408-foot homer to center.
Busch's 367-foot homer to left-center field just carried over the basket to make it 6-1.
