He sustained an ankle injury in last Sunday's victory over the Dallas Cowboys and did not practice on Wednesday or Thursday before being ruled out on Friday.
Waddle, who also got poked in the eye, played only 28 snaps and caught just one pass — a 50-yard reception on Miami's opening drive — against Dallas.
Waddle, 25, has 72 receptions for 1,014 yards and four touchdowns in 14 starts this season.
The 2021 first-round pick is the first player in Dolphins history with 1,000 receiving yards in three straight seasons.
The host Ravens (12-3) can clinch the No. 1 seed in the AFC with a victory Sunday afternoon against the Dolphins (11-4).
