The ATP 250 tournament will take place from Jan. 12-17.
Djokovic, 38, said he fondly remembered the reception he received while winning the tournament in 2023.
"It definitely felt like playing at home, that's for sure," the 24-time Grand Slam champion from Serbia said. "The support that I've been getting in the last 10 days, I don't think I've experienced too many times in my life, so thank you so much for everyone for coming out every single match."
The Adelaide International serves as a pivotal tune-up for the Australian Open, where Djokovic is a 10-time champion. The main draw for the latter tournament is Jan. 18, while the men's singles final is tabbed for Feb. 1.
