Open.
Bu, a 23-year-old China native who is ranked 76th in the world, managed just nine winners to 25 from his Greek opponent. However, Tsitsipas committed 33 unforced errors while Bu gave away only 11 points.
Bu advances to face Argentina's Mariano Navone, who got past 16th-seeded Marcos Giron 6-2, 6-2.
Second-seeded Tallon Griekspoor of the Netherlands also stumbled in the second round as Hungary's Marton Fucsovics posted a 6-3, 4-6, 6-3 win. Third-seeded Luciano Darderi of Italy squandered a match point in the second-set tiebreaker before eventually topping Mackenzie McDonald 7-6 (5), 6-7 (6), 6-4.
The Netherlands' Botic Van De Zandschulp ousted sixth-seeded Matteo Arnaldi of Italy 6-3, 6-3.
