Smotherman, 28, successfully made weight at 135.5 pounds but fell to the floor after stepping off the scale.
He was carried off the platform and examined by physicians at T-Mobile Arena, but no update has been provided.
The scheduled fight between Smotherman (12-6) and Ricky Turcios (13-5) was canceled.
Two other fighters missed weight -- bantamweight Deiveson Figueiredo and flyweight Alex Perez -- and were fined 25% of their purses. Their bouts will continue as planned.
Saturday night's headliner is a clash between Paddy Pimblett and Justin Gaethje for the interim lightweight title.
